Split Luciferase Technology

Luciferase derived from Pyrearinus termitilluminans (Emerald Luc), is known to emit a brighter and more stable signal than conventional firefly luciferases. The N- and C-terminal domains of luciferase can be separated into 2 fragments, which can re-associate in cells. Utilizing this phenomenon, ProbeX has developed a split Emerald luciferase assay system where the two luciferase fragments spontaneously refold to generate a strong and stable signal when they are brought within proximity of each other (patent filed). This split luciferase complementation assay enables the detection of various types of protein-protein interactions, including GPCRs, with ease and high-sensitivity.
